Одним из его организаторов был известный языковед, автор «Толкового Словаря русского языка» Владимир Даль. В основу фондов музея в разные годы вошли коллекции не только музея Оренбургского края (1924 г.), но и Жетысуйского (Семиреченского) областного музея (1929 г.) и республиканского антирелигиозного музея (1941 г.). Музей размещался в здании Вознесенского Кафедрального собора, являющегося уникальным архитектурным комплексом г.
